Hyperuricemia and Inflammatory Reaction:A Study of Their Roles in Pathogenesis of NAFLD

The Expression of Leptin/PI3-K/PKB in Patients with Non-alcoholic Fatty Liver Disease

    Abstract  Objective  To evaluate the expression of leptin,leptin receptor (OB-R) and the components in phosphatidylinositol 3-kinase/protein kinase B (PI3-K/PKB) signaling pathway in patients with non-alcoholic fatty liver disease (NAFLD).Methods  Totally 30 patients with definite diagnosis of NAFLD treated in our hospital between May 2007 and September 2007 were included in this study,and 30 patients with cholecystic polypus but normal liver treated in our hospital were taken as the control group.The method of immunohistochemstry were used to assess the expression of hepatic leptin,OB-R,PI3-K (p85α) and PKB in all participants,and serum leptin level was determined by radioimmunoassay.This study also analyzed whether the expression of these proteins was correlated with each others.Results  The serum leptin level in NAFLD group were significantly higher than that in the control group(10.90±3.39)μg/L vs.(5.55±1.96)μg/L,t=-7.841,P0.05.Compared to the control group,the expression of hepatic leptin,OB-R and PI3-K was also significant higher in NAFLD group (P0.05),while the expression of PKB was significantly lower in NAFLD group (P0.05).In NAFLD group,the expression of hepatic leptin was positively correlated with serum leptin level and the expression of PI3-K(p85α) (r=0.413P0.05;r=0.365P0.05);While the expression of PKB was negatively correlated with serum leptin level and the expression of PI3-K(p85α)(r=-0.923,P0.05;r=-0.854P0.05).In the control group,the expression of hepatic leptin was also positively correlated with serum leptin level and the expression of PI3-K(p85α) (r=0.593,P0.05;r=0.795P0.05).But the expression of PKB was positively correlated with serum leptin level and the expression of PI3-K(p85α)(r=0.686P0.05;r=0.840P0.05).Conclusion  The expression of hepatic leptin and leptin receptor was high in NAFLD patients,but the expression of PKB was relatively low.Over expression of hepatic leptin in NAFLD patients seems lead to leptin resistance instead of activation of PKB,and the impaired activation of PI3-K by leptin and PI3-K might be a new mechanism of hepatic leptin resistance in NAFLD patients.

Related Influencing Factors of T2DM Combined with NAFLD and Its Relationship to Carotid Artery Atherosclerosis

    Abstract  Objective  To explore the related influencing factors of type 2 diabetes mellitus (T2DM) combined with nonalcoholic fatty liver disease (NAFLD) and its relationship to carotid artery atherosclerosis (CAA).Methods  A total of 278 T2DM patients hospitalized in this hospital in 2012 were divided,based on whether combined with NAFLD,into non-NAFLD group (n=152),NAFLD group (n=126).Height,weight,waist circumference (WC),hip circumference (HC),blood pressure (BP),disease duration and other general data were recorded,levels of liver enzymes and glucolipid metabolism detected,and carotid atherosclerotic plaques (CAP) determined.Results  No significant difference in gender,age,BP between 2 groups (P0.05).The body mass index (BMI),waist-hip ratio (WHR),alanine aminotransferase (LT),aspartate aminotransferase (AST),triglyceride (TG),insulin resistance index (HOMA-IR),carotid intima-media thickness (IMT),incidence of CAA were higher,high density lipoprotein cholesterol (HDL-C) lower in NAFLD group than in non-NAFLD group,the difference was significant (P0.05).Partial correlation analysis showed that IMT was positively correlated with HOMA-IR (r=0.368,P0.05).Stepwise Logistic regression analysis showed that T2DM combined with NAFLD and HOMA-IR were risk factors of CAA (P0.05);BMI,TG,WHR,ALT,HOMA-IR were independent risk factors of NAFLD (P0.05).Conclusion  Dyslipidemia,insulin resistance and CAA are more vulnerable to T2DM patients combined with NAFLD.NAFLD is related to BMI,TG,WHR,ALT,HOMA-IR.

Nonalcoholic Fatty Liver Disease and SNPs-11377C/G in Promoter Region of Adiponectin Gene:A Correlation Study in Han Elderly Population in Guilin

Relationship of Prediabetic State to Interaction between Chymase and Traditional Cardiovascular Risk Factors
